Understanding and Treating Malignant Melanoma
Malignant melanoma is a paradigm of aggressive cancers of dismal prognosis. Using tissue biopsies, cellular systems and animal models, we have uncovered unexpected roles of endolysosomal pathways in melanoma initiation, progression and drug response. Here we will show functional evidence supporting the concept of endosomal proteins as oncogenic rheostats with melanoma-restricted activities. In addition we will describe a therapeutic strategy to exacerbate endocytosis and promote an effective tumor self-degradation in vivo. Specifically, we will discuss antitumoral features of dsRNA-based nanocomplexes and the efforts to translate these agents into the clinic by the creation of the biotechnology company Bioncotech Therapeutics.
